چکیده

At least four genetically distinct classes of mutations give rise to triazolealanine resistance in SaZmoneZZa typhimurium. Each class studied affects regulation of the histidine operon. Mutants of one class, GO, are located at one end of the histidine operon. This class may be similar to operator constitutive mutants studied in other systems. At least two genetic sites are present in this region. A second class, hid’, is linked in transduction tests to the guaA and s&B loci. This class contains a low specific activity of histidyl-tRNA synthetase and may represent mutations in the structural gene for this enzyme (Roth & Ames, 1966). Mutations of a third class, hi.sR, are linked in transduction tests to the ilv and m&E loci (Roth & Hartman, 1965) and affect the levels of effective transfer RNA(his) (Silbert, Fink & Ames, 1966). A fourth class of mutants, hisT, is linked to the pzcrB’ and aroD loci.

Histidine Regulatory Mutants in Salmonella typhimurium

Regulation of the histidine operon in Salmonella typhimurium has been shown to involve at least six separate genes (Roth, Antdn & Hartman, 1966; Ant&, 1968). Mutations in any one of these separate genes can destroy the ability of the cell to repress fully the histidine biosynthetic enzymes. In order to understand this regulatory mechanism and the nature of these mutations, we have studied the d...

Isolation of the rec mutants in Staphylococcus aureus.

A histidine auxotroph (his-) of Staphylococcus aureus MS3937 and mutants sensitive to ultraviolet (UV) irradiation were obtained. The UV-sensitive mutants were found also to be sensitive to N-methyl-N'-nitro-N-nitrosoguanidine and mitomycin C, and their sensitivity was accounted for by a defect in deoxyribonucleic acid repair. Transduction of either chromosomal or plasmid markers to UV-sensitiv...

Histidine regulation in Salmonella typhimurium. XV. Procedure for the selection of mutants unable to derepress the histidine operon.

A general search has been made for mutants defective in their ability to derepress the histidine operon. The procedure was to select for mutants with an increased sensitivity to the false feedback inhibitor, 2-thiazolealanine. Five mutant strains defective in derepression have been isolated. All five strains are unable to derepress normally because of mutations located in the operator-promoter ...

Histidine regulatory mutants in Salmonella typhimurium II. Histidine regulatory mutants having altered histidyl-tRNA synthetase.

(1) The hid’ gene of Salmonelb ty(phinzurium is the structural gene for histidyltRNA synthetase. Mutants in the hid gene have an altered histidyl-tRNA synthetase and constitute one of the four classes of histidine regulatory mutants. (2) Mutant hisS1520 has a synthetase with a 60.fold decreased affinity for histidine. This mutant has a markedly decreased growth rate on minimal medium and is de-...

The genetic control of the enzymes of histidine biosynthesis in Salmonella typhimurium.

The last four enzymes in the pathway of L-histidine biosynthesis have been assayed in various histidineless mutants of Salmonella typhimutium. Three classes of mutants (B, C and D) have been shown to be associated with lack of imidazoleglycerol phosphate dehydrase, imidazoleacetol phosphate transaminase, and histidinol dehydrogenase, respectively. These studies, in conjunction with the genetic ...
